Smash-Up 2, and refrain from telling me all the comments that indicate impediments from this game ever existing, I am well aware of them, but I nonetheless wish for it. Also a mature game based on the comics. Both Wii U exclusives! (Again, I am aware that that won't be the case, I am simply wishing.)

I'd like to see a TMNT: Smash Up Melee, lol.
The models should all be from the Nick Cartoon, but the gameplay be like the previous Smash-Up (perhaps faster-paced)-
I really like that free-roaming, platform-oriented, environment-abusing game mechanic for a TMNT fighting game.
That's as far as a TMNT fighting game goes.

For a more regular TMNT game, I also agree with Axtlar. Something that goes the completely opposite way of the cutesy nick cartoons, and goes into a more darker, perhaps even Madworld-style (except instead of pure black and white, we get grayscales with red for blood).
A game that dives me headfirst into the TMNT COMICS universe. With places to explore above the ground or beneath the sewers. Beat'em ups with RPG elements and lots of collectibles. (Items, characters, weapons, you name it). Heck, we could even go to Usagi Yojinbo's dimension every once in a while too.

I would like to see an Arkham Asylum style game. Running through New York with four different turtles with their unique fighting style? That'd be awesome. Also, make it co-op so that people can work together with the four turtles. It would just remind me more of the comics.
Of course, we all want to see another TMNT: Smash-Up. With the new Super Smash coming around, though, I'm not sure if we will see one until after that. But, that's pure speculation by me, nothing more. lol.

Yeah, I agree with both Shihe and Red Turtle, those are exactly the kind of games I want to see! and if they make Smash Up 2, add a lot of characters missing from the first:Baxter Stockman, Triceraton, Savanti Romero, Nobody, Rat King, Leatherhead, Renet, Drako, The Daimyo, Hun, all the Foot Ninja variations (Mystics, robots, etc.), heck, I would even accept Bebop and Rocksteady, I can't say the same for Fishface and Dogpound, though.

My favorite post-classic Turtles games (other than the obvious one) were the 2k3 cartoon based Teenage Mutant Ninja Turtles (GBA version) and the 2007 film's GBA game version. If the new game is in any way like one of those, then I'll be really happy. Of course, it doesn't HAVE to be a 2D sidescroller. It could be 3D, but since I'm a bit tired of city tropes in videogames, I'd want the 3D game to have some sort of distinctive art style. Example 1: Monochromatic, but with a specific hue depending on environment, time of day, etc. So it feels a bit... "acid". That's the graphical style I'd go for in a 3D TMNT game to make it feel less like a generic brawler set in the city. Example 2: This web-game left me a great impression. It was fun, and it was visually appealing. The mechanics were fun, though sometimes glitch though. But the important factor is, that I loved the night-club like neon color scheme.
